Gabapentin binds to α2δ-1 subunits of Calcium channels decreases the release of glutamate and Substance-P neurotransmitters. Nortriptyline inhibits the reuptake of serotonin and norepinephrine by the presynaptic neuronal membrane, thereby increasing the concentration of those neurotransmitters in the synapse.
